REVIEW Ketorolac tromethamine, (±)-5-benzyol-2,3-dihydro-lH-pyrrolizine-1-carboxylic acid, 2-amino-2-(hydroxymethyl)-l,3-propanediol, is a heterocyclic acetic acid derivative. Like other NSAIDs, it decreases prostaglandin synthesis by non-selective competitive inhibition of cyclo-oxygenase (COX-1 and COX-2), producing peripherally-mediated analgesia. Ketorolac is marketed as a racemic mixture of S- and R-enantiomers, but only the S-enantiomer has pharmacologic activity.
